How can I verify the size of my PCR product using a DNA ladder?

You can estimate the size of your PCR product by running it with a DNA ladder on an agarose gel. A DNA ladder contains fragments of known sizes. After running the gel, compare the band from your PCR sample with the bands in the ladder. This tells you how many base pairs long your product is. The bands that travel farther are smaller, while larger ones stay near the top.

For example, if your band matches with the 1,000 bp band on the ladder, your PCR product is about 1,000 base pairs long. It’s important to pick a ladder that includes the size range you are expecting. Use proper gel concentration too. For smaller products, a higher percentage gel gives clearer separation.
